Application:

5-bromo-2-ethoxy-pyridine is a pyridine compound that can be used as an organic synthesis intermediate and pharmaceutical intermediate, mainly used in laboratory research and development processes and chemical production processes.

Preparation:

(1) Add 47g2, 5-dibromopyridine, 8g solid sodium hydroxide and 200ml ethanol into a 500ml reaction bottle, stir and start to 

heat up to the reflux state, the reaction time is 5 hours, stop the reaction, steam out most of the methanol, cool down, add 100ml water, extract the dichloromethane once, and remove the dichloromethane to obtain crude products. The crude product 

was distilled under vacuum to obtain 5-bromo-2-ethoxypyridine with a yield of 98%.

(2) Use 2, 5‑ dibromo-pyridine to treat the newly prepared sodium ethanolin solution in ethanol and heat reflux for 28 hours. The compound 5-bromo-2-ethoxypyridine (3 g, 70%) was obtained by concentrating the mixture and partially placing it in dichloromethane and water, washing the organic layer with salt water, drying with magnesium sulfate, filtering and evaporating under a vacuum strip. LCMS: 202.0[M+1]+

(3) 5-bromo-2-ethoxypyridine. Sodium hydride (14mmol, 330mg) was slowly added to ethanol (15ml) at 0℃. Stir the reactant for 30 minutes and add 5-bromo-2-chloropyrimidine (3.2mmol, 620mg). The reaction mixture was allowed to warm to room temperature overnight, then quenched with water and extracted with ethyl acetate. Combined with the organic layer, the required bromide (470mg, 72%) was obtained by vacuum concentration. MS(M+2)+203.4.
