Steps:
1.3.1 Method 1:
To a solution of NaOH (2.70kg, 4.01eq.) dissolved in water (25.36L, 5V) at 20°C was added 2-(4-(benzyloxy)-3-bromophenyl)acetic acid Methyl ester (5.64 kg, 1.0 eq.). Methyl 2-(4-(benzyloxy)-3-bromophenyl)acetate was dissolved with stirring at 50°C. At 50°C under nitrogen, 8-hydroxyquinolinone (592 g, 0.1 eq.) was added. The reaction mixture was refluxed for 48 hours. After cooling to room temperature, the solids were removed by filtration and the pH of the filtrate was adjusted to 2-3 by concentrated HCl. The solids were collected by centrifugation, the filter cake was transferred to the reactor, and toluene (4V) was added. The mixture was stirred at 70°C until the filter cake dissolved. Stirring was stopped and the mixture was allowed to stand to separate the layers. The organic phase was cooled to 15°C and stirred for 10 h. The resulting solid was collected by centrifugation and the filter cake was dried to give the desired product (3.38 Kg, yellow solid). (78% yield, 85% HPLC purity).
