This thiol-reactive reagent (FW = 139.15 g/mol; CAS 21746-40-7), also
known as NPM and 1-propyl-1H-pyrrole-2,5-dione, readily alkylates
protein sulfhydryl groups. Under alkaline conditions it can also react with
amino groups. Some enzymes are inactivated at faster rates with analogues
having longer alkyl chains (e.g., the inactivation rate of yeast alcohol
dehydrogenase with N-octylmaleimide is significantly faster than with N-
