米氮平杂质E性质、用途与生产工艺
4-甲基-1-(3-甲基-2-吡啶基)-2-苯基哌嗪即米氮平杂质K,为米氮平生产过程中的副产物,反应过程如下:
i. CH3SO2Cl, CH2Cl2, reflux;
ii. LiAlH4, THF, reflux.具体反应步骤为:在N
2气氛下,在0-5℃下,将甲磺酰氯(4.86g,42.45mmol)加入米氮醇(10.0g,35.34mmol),三乙胺(5.35g,52.97mmol)的二氯甲烷(60mL)溶液中,并在25-30℃下搅拌12小时。此后,加热回流2小时。将物质冷却至25-30℃,用水(20mL)洗涤,蒸除溶剂,得到甲磺酰基衍生物中间体(10.22g,80%)。将该残余物不经纯化溶于THF(100mL),并在N
2气氛下在0-5℃下加入LiAlH 4(5.37g,141.32mmol)。 将反应物料加热回流15小时,在0℃用水(5.4mL)、15%NaOH(5.4mL)和水(16.2mL)淬灭,过滤,将滤液蒸干并加入二异丙醚(40mL)。过滤产物用二异丙基醚(10mL)洗涤,干燥,得到呈白色固体的米氮平杂质K(5.66g,75%),纯度99.1%。
