去氟甲基丙酸氟替卡松二硫化物为丙酸氟替卡松的合成中间体,主要用作医药中间体,用于实验室研发过程和化工医药合成过程中。
去氟甲基丙酸氟替卡松二硫化物为丙酸氟替卡松制备过程中生成的杂质:第一步涉及在氟米松中C-17上的羟甲基的氧化裂解,化学上称为6α,9α-二氟-11β,17α,21-三羟基-16。α-甲基-雄甾-1,4-二烯-3,20-二酮,通过高碘酸得到6α,9α-二氟-11β,17α-二羟基-16α-甲基-雄甾-1,4-二烯-3-酮-17β-羧酸2.使用N,N'-羰基二咪唑(CDI)在二甲基甲酰胺(DMF)中活化化合物2的羧基随后用H2S处理得到6α,9α-二氟-11β,1-7α-二羟基-16α-甲基-3-氧代-雄甾-1,4-二烯-17使用丙酰氯和三乙胺(TEA)将化合物3的C-17羟基酯化,得到6α,9α-二氟-11β-羟基-16[甲基]-[化学式3]的C-17羟基。α-甲基-17α-丙酰氧基-3-氧代-雄甾-1,4-二烯-17β-硫代甲酸4.使用NaHCO3和二甲基乙酰胺将化合物4与溴氯甲烷烷基化ide(DMAc)得到S-氯甲基-6α,9α-二氟-11β-羟基-16α-甲基-17α-丙酰氧基-3-氧代-雄甾-1,4-二烯-17β-硫代磷酸酯。
