4-安替比林甲醛性质、用途与生产工艺
安替比林又名非那宗。无色或白色结晶性粉末,无臭,味微苦。熔点110—113℃。易溶于水、乙醇、氯仿,略溶于乙醚。由苯肼与丁酮酰胺环合后,再与硫酸二甲酯进行甲基化反应制得。为解热药物,作用迅速,持续时间较长,具有微弱的镇痛、镇痉作用。安替比林(PZ)是一种广泛使用的消炎镇痛药物,研究人员在德国Berlin地下水和饮用水中检出安替比林以及该药物的代谢物。
研究表明安替比林对肺及黏膜具有毒性,长期暴露将会导致靶器官受损,对人健康造成危害,4-安替比林甲醛为安替比林衍生物,可用作医药合成中间体。
4-安替比林甲醛制备如下:
(1)将安替比林加到配制罐,再加入配量硫酸和水,配成含量为34-36%的酸性溶液,然后与亚硝酸钠溶液在亚硝化小桶中进行亚硝化反应,反应液放入还原罐中,加入还原剂进行还原反应,再将还原液转入水解罐,加入硫酸水解后,通入氨气中和,制得4-氨基安替比林油;
(2)将4-氨基安替比林油加入到酰化罐中与甲酸甲酯或甲酸进行酰化反应,然后降温结晶离心得到4-安替比林甲醛结晶。
