背景

肿瘤坏死因子受体家族包括TNF-RI、FAS、DR3、DR4、DR5和DR6,在各个生理系统中调控细胞凋亡方面起着重要作用。受体由细胞因子家族激活,该家族包括TNF、FasL和TRAIL。它们的特点是含有一个高度保守、富含半胱氨酸的重复序列的胞外区和一个胞内保守的约80个氨基酸的死亡结构域(DD)通过募集其他含有DD的接头蛋白(FADD、TRADD、RIP)至死亡诱导信号转导复合体(DISC),并进而导致caspase激活,DD在转导死亡信号中发挥重要作用。
凋亡受体信号转导还受诱饵受体家族(DcR1、DcR2和DcR3)控制,这些诱饵受体缺少胞质DD并且通过对配体竞争而抑制死亡受体介导的凋亡。诱饵受体的表达提供了某些类型癌症调节凋亡的机制并且可能有助于化疗敏感性。

应用

ANTI-DCR1用于TRAIL及其受体DR5、DcR1在初发和复发尖锐湿疣患者皮损中的表达及意义的研究。
