苯酚 - 氯仿 - 异戊醇混合物性质、用途与生产工艺
苯酚-氯仿-异戊醇混合物应用举例如下:
1)用于一种适用于全基因组测序的产糖被细菌基因组DNA的提取,向产糖被细菌的培养物中加入提取缓冲液和溶菌酶,37℃下静置10min,再加入蛋白酶K,65℃下静置1h;加入苯酚-氯仿-异戊醇的混合溶液,抽提;加入氯仿-异戊醇混合溶液,抽提;加入冷乙醇和醋酸钠,离心,沉淀经乙醇洗涤后室温干燥,加DNA提取缓冲液溶解;加入RNA酶,37℃下静置10min;加入PEG6000,混匀,50℃下静置10min;加入苯酚-氯仿-异戊醇的混合溶液,抽提;加入氯仿-异戊醇混合溶液,抽提;加入冷乙醇和醋酸钠溶液,离心,沉淀经乙醇洗涤后室温干燥,加ddH2O或者TE缓冲液溶解,即为提取到的DNA溶液。
苯酚-氯仿-异戊醇混合物常用作分析提取液用于提取DNA、多糖等。
