N-芴甲氧羰基-N'-生物素-L-赖氨酸性质、用途与生产工艺
N-芴甲氧羰基-N'-生物素-L-赖氨酸又叫Nα‑芴甲氧羰基‑Nε‑生物素‑赖氨酸,是一种常用的氨基酸保护试剂,在多肽合成领域经常用到。
将1当量的Nα‑芴甲氧羰基‑Nε‑叔丁氧羰基‑L‑赖氨酸溶解于二氯甲烷中,加入1份的质量百分浓度为15%三氟醋酸的二氯甲烷溶液,剧烈搅拌,外部用自来水冷却,薄层色谱检测反应完全,减压蒸馏掉溶剂和三氟醋酸,加入二氯甲烷再次溶解,再次减压蒸馏,石油醚洗涤,石油醚倾掉,将剩余固体溶于二甲基甲酰胺中,加入2当量N‑甲基吗啉,pH值调到8,缓慢加入0.7当量的N‑羟基琥珀酰亚胺生物素,反应3小时,薄层色谱显示反应完全,向反应中加入柠檬酸溶液,pH调至3,固体析出,洗涤,烘干,得产品,纯度99.1%,收率90%。
