VEGF, a homodimeric glycoprotein of relative molecular mass 45,000, is the only mitogen that specifically acts on endothelial cells. It may be a major regulator of tumor angiogenesis in vivo. Vascular endothelial growth factor is a mitogen primarily for vascular endothelial cells. It is, however, structurally related to platelet-derived growth factor. VEGF shares homology with the PDGF A chain and B chain, including conservation of all 8 cysteines found in PDGFA and PDGFB. VEGF gene contains 8 exons. Vascular endothelial growth factor (VEGF) induces remodeling and enhances TH2-mediated sensitization and inflammation in the lung. 血管内皮生长因子(vascular endothelial growth factor,VEGF),又称血管通透因子(vascular permeability factor,VPF)是一种高度特异性的促血管内皮细胞生长因子,具有促进血管通透性增加、细胞外基质变性、血管内皮细胞迁移、增殖和血管形成等作用。
血管内皮生长因子是一个家族,包括VEGF-A、VEGF-B、VEGF-C、VEGF-D、VEGF-E和胎盘生长因子(PGF)。通常VEGF即VEGF-A。VEGF-A可促进新生血管形成和使血管通透性增加,VEGF-B在非新生血管形成的肿瘤中起作用,VEGF-C和VEGF-D在癌组织的新生血管和新生淋巴管的形成过程中起作用,VEGF-E也是一种潜在的新生血管形成因子,PGF促进新生血管形成,使血管通透性增加,在实验性脉络膜新生血管中PGF的表达明显增高。
