Uses | Anti-Sirt7 antibody produced in rabbit has been used in western blotting analysis and immunofluorescence detection. | General Description | This gene encodes a member of the sirtuin family of proteins, homologs to the yeast Sir2 protein. Members of the sirtuin family are characterized by a sirtuin core domain and grouped into four classes. The functions of human sirtuins have not yet been determined; however, yeast sirtuin proteins are known to regulate epigenetic gene silencing and suppress recombination of rDNA. Studies suggest that the human sirtuins may function as intracellular regulatory proteins with mono-ADP-ribosyltransferase activity. The protein encoded by this gene is included in class IV of the sirtuin family. (provided by RefSeq) | Biochem/physiol Actions | Sirt7 (sirtuin 7) is extensively found in protein associated with active rRNA genes (rDNA), in the nucleolus. It interacts with RNA polymerase I (Pol I) and histones. Overexpression of Sirt7 increases Pol I-mediated transcription, whereas knockdown of Sirt7 or inhibition of its catalytic activity results in decreased association of Pol I with rDNA and reduces Pol I transcription. Depletion of Sirt7 stops cell proliferation and triggers apoptosis. High levels of Sirt7 expression are associated with breast cancer. |
