Raltetrexed is a quinazoline folate analogue, whose anti-tumor effect is generated by the specific inhibition of thymidine synthase. In the treatment of colorectal cancer, its efficacy is similar to that of fluorouracil, but its toxicity and side effects are smaller than that of fluorouracil, and its administration method is relatively simple. Antitumor agents, drugs affecting nucleic acid biosynthesis, used for: ① advanced rectal and colon cancer. ② Breast cancer, ovarian cancer, non-small cell lung cancer, pancreatic cancer, hepatocellular carcinoma. Clinical trial is now in: ① solid tumor: combined with taxane drugs such as taxol, Taxantel. ② Non-small cell lung cancer: combined with paclitaxel and carboplatin. ③ Locally advanced or metastatic gastric cancer: combination of doxorubicin, daunorubicin and other anthracycline drugs. ④ Metastatic colon and rectal cancer: combined with oxaliplatin and other platinum drugs. ⑤ Rectal cancer: combined with linentine and other topoisomerase inhibitors. Colorectal cancer: combined with fluorouracil. Colorectal cancer is a common cancer in western countries. The success rate of surgical treatment is based on the progression of the disease, and chemotherapy is often required after surgery. In the past 35 years, fluorouracil has been the most common chemotherapy agent. However, fluorouracil has severe toxicity and complex administration regimen, while raltetrexel has similar anti-tumor effects to fluorouracil, but less adverse reactions. Therefore, fluorouracil can be replaced in the treatment of advanced colorectal cancer.
