Appearance: white acicular crystal. Melting point 68-69℃; Boiling point 180-182℃ (0.53kPa), 159-160℃ (0.267kPa) soluble in ethanol, ether, benzene, chloroform and oil, insoluble in water. Commonly used base hydrochloride, lidocaine hydrochloride (C14H22N2O·HCL, [73-78-9]) is a white crystalline powder. The melting point is 127-129℃, and the melting point of monohydrate is 77-78℃. Highly soluble in water, pHO 4.0-5.5 in 0.5% aqueous solution. Odorless and bitter.

Use 1

The product is an amide local anesthetic. It is widely used in surface anesthesia, infiltration anesthesia, conduction anesthesia and epidural anesthesia. The oral LD50 of lidocaine hydrochloride in mice is 290mg/kg.

Method one

It is obtained by acylation and amination of 2, 6-dimethylaniline. 1. Acylation dissolve 2, 6-dimethylaniline in anhydrous benzene, cool to below 28℃, stir slowly add chloro-acetyl chloride, at a controlled temperature below 30℃. After stirring reaction for 1h, reheating reflux for 8h. Crystallized by cooling, filtered and dried, 2, 6-dimethylchloroacetanilide was obtained. The yield is 75%. 2. Amination Add the above 2, 6-dimethylchloroacetanilide to benzene, then add diethylamine, stir and heat reflux for 7h. After recovering most of the benzene, the crystals are cooled, filtered, and washed with benzene. Benzene solution was combined and extracted with 10% hydrochloric acid. The extraction solution was decolorized with activated carbon and filtered. The filtrate was adjusted to pH 10 with 10% sodium hydroxide solution, crystallized, filtered, washed to neutral, and lidocaine. After recrystallization, the salt from hydrochloric acid is lidocaine hydrochloride. The yield of amination was 76%.
