Pharmacology and toxicology
Changchun alkali is a kind of alkaloid that has anticancer activity. It mainly inhibits the polymerization of microtubules and prevents the formation of microtubules of the spindle, which stops mitosis in the medium term. It can also be applied to cell membrane, interfering with the transport of amino acids by cell membrane, which can inhibit protein synthesis and inhibit RNA synthesis. Vinblastine antineoplastic spectrum broad, in 0.05 ~ 0.1 mg/kg doses in mice L1210 leukemia, airy, S180 ascites carcinoma, C3H mice spontaneous and metastatic breast cancer have obvious anti-tumor activity, to transplantation in rat cheek pouch of wool also has inhibitory effect on the growth of cancer cells.
pharmacokinetic
Oral absorption is poor and requires intravenous administration. After the injection of changchun base, it was rapidly distributed in various tissues, rarely penetrating the blood-brain barrier, and the protein binding rate was 75%. Blood was used to remove the drug in two directions, with t1/2 alpha for 4.5 minutes, t1/2 beta for 190 minutes, and the tip elimination phase t1/2 gamma for 24 hours. In the liver, most of it was excreted with bile, and 33% of the drug was excreted in the feces within 3 days after use, mainly metabolites, and 21 percent were excreted with urine.
indications
Mainly used for the treatment of solid tumors. The efficacy of malignant lymphoma, testicular tumor and choriocarcinoma is better, and it also has some effect on lung cancer, breast cancer, ovarian cancer, skin cancer, renal cell tumor and monocyte leukemia.
Hodgkin's disease and malignant lymphoma can also be used to treat chorionic epithelial carcinoma and breast cancer.
