Uses

Hepatocellular Carcinoma (HCC):

Sorafenib is primarily used to treat hepatocellular carcinoma, which is the most common type of liver cancer. It is usually prescribed for patients with advanced HCC who are not candidates for surgical resection or liver transplantation.

Renal Cell Carcinoma (RCC):

Sorafenib is also approved for the treatment of advanced renal cell carcinoma, a type of kidney cancer. It is used in patients who have failed prior systemic therapy or for those who are not suitable for such treatments.

Thyroid Cancer:

Sorafenib is used to treat differentiated thyroid carcinoma (DTC) that is refractory to radioactive iodine treatment. This includes papillary, follicular, and Hürthle cell thyroid cancers.

Mechanism of Action

Tyrosine Kinase Inhibition: Sorafenib works by inhibiting multiple tyrosine kinases, which are enzymes involved in the signaling pathways that regulate cell division and growth. By blocking these enzymes, Sorafenib interferes with the growth and proliferation of cancer cells.

Anti-Angiogenesis: Sorafenib also inhibits vascular endothelial growth factor receptors (VEGFR) and platelet-derived growth factor receptors (PDGFR), which are involved in the formation of new blood vessels (angiogenesis). By blocking these receptors, Sorafenib reduces the blood supply to tumors, thereby inhibiting their growth.

Precautions

Liver Function: Sorafenib can cause hepatotoxicity, so liver function should be monitored regularly. Dose adjustments may be necessary based on liver function tests.

Cardiovascular Health: Sorafenib can cause hypertension (high blood pressure) and increase the risk of cardiac events. Blood pressure should be monitored regularly, and any cardiac symptoms should be reported immediately.
