1. 原理与应用
高碘酸钠氧化法活化辣根过氧化物酶（HRP）的糖基，经过活化的HRP带有醛基可以与待标记物的游离氨基结合，发生希夫碱反应（Schiff's base reaction）。本试剂盒适用于带有游离氨基的物质标记，如：带有游离氨基（伯胺）的抗体、蛋白或者小分子化合物。

3. 缓冲液配置
(1) 0.01M CB，PH:9.6缓冲液配制：取NaHCO3, 0.67g和Na2CO3, 0.21g加水到1L溶解，不用调节PH。
(2) 0.067M PBS, PH:7.0缓冲液配制：取NaH2PO4.2H2O，5.35g和Na2HPO4.12H2O, 11.7g以及NaCL，9g加水1L溶解，添加0.35g NaOH再次混匀，不用调节PH。
4. 使用方法
(1) 首先去除待标记抗体或蛋白中的叠氮钠、以及含有氨基的缓冲成分物质，比如甘氨酸、Tris 等，还有EDTA等物质，可采用透析和超滤的办法置换缓冲液，以去除干扰物质达到标记效果！透析或超滤缓冲液首选0.01M CB,PH:9.6缓冲液，若实验室没有碳酸盐试剂再采用0.01M PBS（PH:7.0-7.4均可）透析或超滤置换缓冲液，最后调整抗体或蛋白的浓度到2MG/ML。如果抗体和蛋白不含上述这些干扰物质可以直接用上述PBS或CB缓冲液调整浓度到2MG/ML备用。
(2) 从-20℃取出HRP标记试剂盒，室温条件25-30℃温箱平衡30分钟左右，使各组分充分解冻后混匀。
(3) 取500uL抗体或蛋白（约1mg），加入一只预活化HRP(1mg)冻干粉中，用移液枪反复吹打几次混匀HRP和抗体，混匀后加入HRP标记启动液80ul继续混匀数次，避免产生气泡。最后室温避光或25-30℃温箱偶联反应2-3小时，若遇快要下班也可放4℃反应24小时左右（效果一样或者更好不必担心）。
(4) 偶联结束将反应终止液粉管中加入1mL去离子水混匀，取50ul加入3所述反应液中，充分混匀，室温放置1小时或者4℃ 2小时。
(5) 终止完成后，可直接加入等体积的标记物保存液，充分混匀，置于-20℃保存。如果长期保存,建议先用0.067M PBS PH=7.0透析或超滤置换缓冲液后再加标记物保存液。可以保存3年以上。
