背景(Background)
受体酪氨酸蛋白激酶erbB-4(ErbB4),也称为Her4,是一种单程I型跨膜糖蛋白,是酪氨酸激酶受体erbB家族的成员。ErbB家族成员是表皮生长因子(EGF)家族生长因子的受体。ErbB4在正常骨骼肌、心脏、垂体、脑和几种乳腺癌中表达。ERBB4包含多个富含半胱氨酸的弗林蛋白酶样结构域、一个酪氨酸激酶结构域、磷酸肌醇-3激酶结合位点和一个PDZ结构域结合基序。该蛋白与神经调节蛋白-2和-3、肝素结合EGF样生长因子和β细胞蛋白结合并被其激活。配体结合诱导多种细胞反应,包括有丝分裂和分化。多种蛋白水解事件允许释放细胞质片段和细胞外片段。ErbB4在神经元发育、心脏发育和癌症中起着重要作用。ERBB4已被证明与DLG4、NRG1、STAT5A和YAP1相互作用。该基因的突变与癌症有关。其他单核苷酸多态性和风险单倍型与精神分裂症有关。
