背景（Background）
CD40配体也称为CD40L、CD154、TNFSF5和T细胞抗原Gp39，是一种单程I型膜蛋白，属于TNF超家族分子。CD40配体主要在活化的CD4+T淋巴细胞上表达，也存在于其他类型的细胞中，包括血小板、肥大细胞、巨噬细胞、嗜碱性粒细胞、NK细胞、B淋巴细胞以及非造血细胞（平滑肌细胞、内皮细胞和上皮细胞）。尽管可溶性CD40配体的所有单体、二聚体和三聚体形式都可以与CD40结合，但三聚体形式的可溶性CD40配位体通过细胞表面CD40的寡聚化具有最强的生物活性，这是TNF受体家族成员的共同特征。
CD40配体与抗原呈递细胞（APC）上的CD40结合，这会根据靶细胞类型产生许多影响。一般来说，CD40配体起着共刺激分子的作用，与APC上MHC分子对T细胞受体的刺激相关，诱导APC的激活。CD40配体共有三个结合伴侣：CD40、α5β1整合素和αIIbβ3。CD40配体通过与B细胞表面的CD40结合来调节B细胞功能。该基因的缺陷导致无法进行免疫球蛋白类转换，并与高IgM综合征有关。
