According to Scheme 5 Step 1 : A solution of l-(2-chloro-4-nitrophenyl)piperazine (2.90 mmol, 700 mg), BoC2O (3.19 mmol, 0.96 mL), Et3N (3.48 mmol, 0.49 mL) and of DMAP (0.29 mmol, 35 mg) in DCM (5 mL) was stirred at room temperature overnight. The organic phase was washed once with HCl 0.1 M, once with water, once with brine, was dried over Na2SO4, was filtered and was concentrated under reduced pressure to afford tert-butyl 4-(2-chloro-4-nitrophenyl)piperazine-l-carboxylate (2.50 mmol, 854 mg, 86%). The product was used without further purification. LC (Zorbax SB-Ci8, 3.5μm, 4.6x30mm Column): RT = 3.19 min; MS m/z ES+= 342.
