Preparation of 4-chloro-6-methoxy-5-nitro-2-(propylthio)pyrimidine (CMLIN)


To a solution of CLIN (1.0 g, 3.73 mmol) in dry MeOH (10 mL) NaOMe (0.20 g, 3.73 mmol) was slowly added, and reaction mixture was then stirred at room temperature for 1 h. AcOH (1 mL) and water (20 mL) were added, and product was extracted to MeTHF (3 x 10 mL).
Combined organic phases were dried over MgSO₄ and concentrated to afford crude product, which was then purified by chromatography (SiO₂, hexane:EtOAc) to afford yellow oil (0.78 g, 79% yield). MS (ESI) m/z: 264 [MH]⁺.
