Reaction Conditions:

Stage #1:indole with potassium hydroxide in dimethyl sulfoxide at 20; for 1 h;Inert atmosphere;
Stage #2:2-bromoethanol in dimethyl sulfoxide at 20; for 18 h;Inert atmosphere;

Steps:

2 Synthesis of 2-indol-1-ylethanol (Intermediate 2) and 2-(2-indol-1-ylethoxy)ethanol (Intermediate 3)
To a suspension of KOH (4.48 g, 80.0 mmol) in DMSO (20 mL) is added R-2 (2.34 g, 20.0 mmol) under N2 and the suspension is stirred at room temperature for 1 h. 2-Bromoethanol (1.8 mL, 25.0 mmol) is then added and stirring continued for 18 h. The reaction is diluted with EtOAc and washed with water, and the organic phase dried over anhydrous Na2SO4. The solvent is removed in vacuo and the crude material purified by flash chromatography (SiO2, 1% MeOH in DCM) to give I-2 (300 mg) m/z 162.0 [M+H] and I-3 (75 mg) m/z 206.0 [M+H].
