Synthesis of 2-(1H-indol-5-yl)acetic acid
NaOH (166 mg, 4.15 mmol) was added to a solution of methyl 2-(1H-indol-5-yl)acetate (393 mg, 2.08 mmol) in THF (6 mL) and H2O (3 mL).
The mixture was stirred at 50°C overnight.
The resulting mixture was concentrated, and water (15 mL) was added.
The mixture was acidified to pH=1 with aqueous HCl (12 M), and DCM (50 mL*3) was added to extract the desired compound.
The combined organic solution was washed with brine (50 mL), dried over anhydrous Na2SO4 and it was concentrated to obtain the desired compound (341 mg, yield 94%) as an ocher solid. MS (ESI) Rt = 1.142 min, m/z 176.0 [M+H]+; Purity: 100% 214 nm.
