Reaction Conditions:

Stage #1: benzyl bromidewith iodine;zinc in N,N-dimethyl-formamide at 15 - 85; for 3 h;Inert atmosphere;
Stage #2: 2-amino-3,5-dibromopyrazinewith bis-triphenylphosphine-palladium(II) chloride in N,N-dimethyl-formamide at 15 - 30; for 12 h;

Steps:

1 Preparation of intermediate 2-amino-5-bromo-3-phenylmethylpyrazine (intermediate 1-1):

Under nitrogen protection, activate zinc powder (5g, 77mmol) and elemental iodine (500mg)Mix, add DMF, and stir at room temperature for 10 minutes until the color of iodine disappears.Then benzyl bromide (3.6ml, 25.6mmol) was added and the oil bath was refluxed at 85°C for 3h.After 3h, move to room temperature, add 2-amino-3,5-dibromopyrazine (5g, 20mmol) and bistriphenylphosphine palladium dichloride (700mg, 1mmol). Reaction at room temperature for 12h. The reaction solution was filtered through celite,After adding ethyl acetate and saturated ammonium chloride solution repeatedly to extract three times, the ethyl acetate phase was dried over anhydrous sodium sulfate, filtered, and purified with 200-300 mesh silica gel to obtain intermediate 1-1 as a yellow solid 4.86g, yield 91.8 %.
