To a solution of 3-methoxyphenol (5.0 g, 40.3 mmol) in CHCl3 (50 mL) at 0° C. was added SO2Cl2 (3.9 mL, 48.3 mmol) slowly. The resulting solution was stirred for 30 min under argon atmosphere, then heated to 65° C. and stirred for 3 h. The reaction mixture was allowed to cool down to room temperature, poured into ice cold water (100 mL) and extracted with CHCl3, washed with saturated NaHCO3 solution and brine. The organic layer was dried over anhydrous magnesium sulfate and concentrated in vacuo. The target compound HJ-3-272 [18113-03-6] was isolated as a colorless oil by flash chromatography on silica gel using EtOAc:Hexane (1:9) as an eluent. Yield: 4.57 g (72%). 1H NMR (300 MHz, CDCl3) δ 7.16 (d, J=8.9 Hz, 1H), 6.61 (d, J=2.9 Hz, 1H), 6.44 (dd, J=8.9, 2.9 Hz, 1H), 6.06 (d, J=1.8 Hz, 1H), 3.73 (s, 3H); 13C NMR (75 MHz, CDCl3) δ 159.6, 152.1, 129.2, 111.6, 107.6, 102.1, 55.4.
