Steps:
To 2-chloroisonicotinic acid Int-Jla (1g, 6.35mmol)Dimethyl sulfoxide (10mL)Add potassium hydroxide (2.00g, 35.64mmol) andEthanol (3.16 g, 68.50 mmol).The reaction mixture was stirred at 80°C for 16 hours.After the completion of the reaction, the reaction mixture was poured into water (100 mL), 1N hydrochloric acid was added to acidify, and the pH of the reaction solution was adjusted to 2. Then the mixture was extracted with ethyl acetate (30 mL×2). The organic phases were combined, washed with water (30mL×2), saturated sodium chloride solution (30mL), dried over sodium sulfate, filtered and concentrated.The title compound Int-J1 (600 mg, yield: 57%) was obtained.
