6 Procedure for Preparation of Compound 5:

Compound 4(1.90g, 9.31mmol) was dissolved in MeOH-DCM(v/v: 2: 1) (20mL). The solution was cooled to 0 °C , and KOH (1.04g, 18.63mmol) was added. The reaction mixture was stirred for 5min at 0 °C , then allowed to warm to RT and stirred for 0.5 h again. TLC showed that the SM was consumed absolutely. 20mL of water was added to the mixture. The mixture was partitioned. The aqueous phase was extracted with DCM (15mLx3). Combine organic phase was washed, dried over anhydrous Na₂SO₄ and concentrated to give benzofuran-2-ylethyne (1.3g,Yield: 98%).
