This illustrates the preparation of (2-Methyl-4-phenyl-l,3-miazol-5-yl)acetic acid. EPO A solution of 3-bromo-4-oxo-4-phenylbutanoic acid (920 mg) and thioacetamide (270 mg) in isopropanol (20 mL) was heated at 80 °C for 3 h. The mixture was evaporated to dryness and the residue was dissolved in a mixture of 4 : 1 : 1 isopropanol / THF / water (20 mL). Sodium hydroxide (1 g) was added portionwise and the mixture was stirred for 1 h. The mixture was concentrated in vacuo and the residue was dissolved in water and extracted with ethyl acetate. The aqueous layer was acidified by the addition of 2 M aqueous hydrochloric acid and then extracted three times with ethyl acetate. The combined organic phases were dried over magnesium sulfate and concentrated in vacuo to give the title compound (480 mg). APCI+MS : m/z 234 [M+H]+.
