10.1 Step 1. Synthesis of 2-morpholinyl-5-nitrobenzonitrile (10a):
2-Bromo-5-nitrobenzonitrile (1.0 g, 4.4 mmol) was dissolved in 20 mL of tetrahydrofuran.Further potassium carbonate (1.22 g, 8.8 mmol) and morpholine (0.58 g, 6.6 mmol) were added.Heat to reflux for 4 h. After the reaction was completed, it was cooled to room temperature and filtered.The filtrate was concentrated and purified by silica gel column chromatography (PE: EA=4:1, v/v).10a, yield 96.3%
