Reaction Conditions:

with 1,1'-bis(diphenylphosphanyl)ferrocene;tris-(dibenzylideneacetone)dipalladium(0);sodium tertiary butoxide in toluene at 100; for 31 h;

Steps:

1.1 (1) Synthesis of intermediate 1-BrNH:

In a 250mL there-necked flask with a magnetic rotor, 2-amino-3-bromopyridine (10.38g, 60.00mmol, 1.00eq) was added successively,Tris(dibenzylideneacetone)dipalladium (549 mg, 0.60 mmol, 1 mol%),1,1'Bis(diphenylphosphine)ferrocene (1.33g, 2.40mmol, 4mol%), sodium tert-butoxide (11.53g, 120mmol, 2.0 equiv), after evacuating nitrogen three times, inject iodobenzene ( 12.24 g, 60.00 mmol, 1.00 equiv) and toluene (150 mL), then stirred in an oil bath at 100 °C for 31 h, cooled to room temperature, and distilled under reduced pressure to remove the solvent to obtain a crude product. The obtained crude product was separated and purified by silica gel column chromatography, and the volume ratio of eluent: petroleum ether/dichloromethane was 10:1-5:1 to obtain 14.62 g of the product 1-BrNH orange-yellow liquid with a yield of 98%.
