1 Example 1: Synthesis of 3-amino-1-benzylpyrrolidine-3-carboxylic acid
1-Benzylpyrrolidin-3-one (8.76 g, 0.05 mol), potassium cyanide (5.86 g, 0.09 mol) and ammonium carbonate (50.7 g, 0.65 mol) were added to 50% ethanol (135 mL) and heated to reflux 3 hours. Ethanol is distilled off under normal pressure, and the residue is cooled and crystallized, and filtered.3-Amino-1-benzylpyrrolidine-3-carbonitrile (8.09 g, 80.4%) was obtained as a brown powder.The compound obtained above and barium hydroxide (63.40 g, 0.37 mol) were added to water (300 mL) under a nitrogen atmosphere and heated to reflux for 36 hours. After cooling to room temperature, water (100 mL) was added and the pH was adjusted to 3 with 6 mol/L sulfuric acid (about 100 mL). Filtration, adding triethylamine (about 40 mL) to the filtrate to adjust to pH=6, standing for crystallizing, filtering, and drying the cake to obtain a brown powdery solid 3-amino-1-benzylpyrrolidine-3-carboxylic acid (7.42 g, 83.8%).
