Reference Example 22; 1-(methylsulfonyl)pyrrolidin-3-amine hydrochloride; [Show Image] To a solution of tert-butyl pyrrolidin-3-ylcarbamate (10.3 g) and triethylamine (9.06 mL) in THF (150 mL) was slowly added methanesulfonyl chloride (6.53 g) at 0°C, and the mixture was stirred overnight at room temperature. To the reaction solution was added ethyl acetate, and the mixture was washed with water and saturated brine, dried over sodium sulfate, and concentrated under reduced pressure. The residue was dissolved in ethyl acetate (250 mL), 4N hydrogen chloride-ethyl acetate solution (50 mL) was added thereto, and the mixture was stirred overnight at room temperature. The solvent was evaporated under reduced pressure to give the title compound (10.03 g, yield 90%) as a white solid. 1H NMR (300 MHz, DMSO-d6) δ: 1.98-2.07 (1 H, m), 2.17-2.27 (1 H, m), 2.97 (3 H, s), 3.27-3.37 (2 H, m), 3.43-3.57 (2 H, m), 3.834 (1 H, br), 8.62 (3 H, br).
