13.3 Step-3: Synthesis of 4-chloro-7-methoxyquinoline-3-carbonitrile:

To a solution of 7-methoxy-4-oxo-1,4-dihydroquinoline-3-carbonitrile (0.1 g, 0.49 mmol, 1 eq) in POCl3 (1 mL) was added DMF (5 drops) and the mixture was allowed to stir at 120 °C for 2 h. Progress of reaction was monitored by TLC. After completion, reaction mixture was cooled to RT, diluted with cold water (100 mL) and filtered. Solid was dried under vacuum to afford 4-chloro-7-methoxyquinoline-3-carbonitrile (60 mg, 60%) as white solid. LCMS: 219 [M+1]+
