Dissolve 3-aminobenzothiophene-2-carboxylic acid ethyl ester (5g, 23mmol), and formamidine acetate (7.2g, 69mmol) in 23mL of ethylene glycol methyl ether,The reaction system was evacuated to change nitrogen, heated to 150°C and refluxed overnight,After TLC detects that the reaction is complete, the system is cooled to room temperature, the product is precipitated, filtered, and the product is washed with ethanol until the filtrate is colorless.The solid was collected and dried to obtain an off-white granular solid benzo[4,5]thiophene[3,2-d]pyrimidin-4(3H)-one (4.38g, 94.1%).
