45.A Step A: Preparation of 4-methyl-2-phenylthiazol-5-amine
Example 45 11 -((3 S,4R)-4-(3,4-difluorophenyl)- 1 -(2-methoxyethyl)pyrrolidin-3-yl)-3-(4-methyl-2- phenylthiazol-5-yl)urea1006521 Step A: Preparation of 4-methyl-2-phenylthiazol-5-amine: N-( 1- cyanoethyl)benzamide (500 mg, 2.87 mmol, prepared as described in Example 41, Step B) and Lawesson’s Reagent (1277 mg, 3.16 mmol) were combined in 2 mL of toluene in a sealed vessel and heated at 100 °C for 2 days. The reaction was cooled, concentrated and purified by silica column chromatography eluting with 10-25% EtOAc-hexanes to provide the title compound (148 mg, 27.1 % yield). MS (apci) mlz = 191.0 (M+H).
