Synthesis of 2-thiol-4-methylquinoline

The initial compounds 2-hydroxy-4-methylquinoline and 2-chloro-4-methylquinoline were synthesized according to themethod reported earlier (Bhojya Naik et al. 2006). Anethanolic solution of 2-chloro-4-methylquinoline was mixedwith sodium sulfide in a 1:3 ratio and refluxed for about 4 hin the presence of a catalytic amount of HCl. The completionof the reaction was monitored by thin-layer chromatography(ethyl acetate:carbon tetrachloride 8:2). Thereaction mixture was poured into ice cold water and neutralizedwith sodium carbonate. The product was filteredand recrystallized. Anal. calc. for C10H9NS: yield 74%, C, 68.51; H, 5.20;N, 7.99. Found: C, 68.57; H, 5.12; N, 8.01; UV-Visible. λmax (nm): 281, 380, IR (KBr) cm-1: 1653 ν(C=N), 2480 ν(C-SH); 1HNMR(δ ppm): 7.1-8.09 (m, ArH, 5H), 2.4 (s,3H, CH3), 13.4 (s, SH), 13C NMR (DMSO-d6) δ: 19.2(CH3),116.3(CH), 124.2, 126.3, 129.2, 145.2, 146.5, 180.6(-C=N), M.W. 175.26.
