Steps:
1.1 Step 1: tert-butyl 4-(5-formylpyridin-2-yl)piperazine-1-carboxylate
Compound 6-bromonicotinaldehyde (1.0 g, 5.376 mmol), N-Boc-piperazine (1.0 g, 5.376 mmol) and DIEA (2.5 mL) were added to 10 mL of DME, and the mixture was warmed to 120 ° C and stirred for 6.5 hours. Water was added, and the mixture was extracted with EA, washed with saturated saline, dried over anhydrous sodium sulfate and concentrated to obtain 1.4 g of a yellow solid (yield: 89.7%).
