5-Methyl-2-(2-nitrophenyl)oxazole synthesis

3synthesis methods
-

Yield:52829-70-6 86%

Reaction Conditions:

Stage #1: 2-nitro-N-(2-oxopropyl)benzamidewith trichlorophosphate at 90; for 0.5 h;
Stage #2: with sodium hydroxide in water;

Steps:

1

To 202 mg (0.91 mmol) of 2-nitro-N-(2-oxopropyl)benzamide was added 1 mL of phosphorus oxychloride. The reaction was stirred at 90 °C for 30 min, then concentrated in vacuo. The residue was taken up in 5 mL of water, then the suspension was extracted with ethyl acetate (3 x 5 mL). The combined ethyl acetate layers were back extracted with 2M NaOH(α(y ) (2 x 5 mL), and brine (1 x 5 mL), dried over MgSO4, filtered, and concentrated in vacuo to a solid. The product was purified via MPLC, eluting with a 20-50% ethyl acetate/ pentane gradient to give 159 mg (86%) of a solid.
