Reaction Conditions:

Stage #1: thiosemicarbazide;3-Phenylpropionic acidwith trichlorophosphate at 75; for 0.5 h;
Stage #2: with water for 4 h;Reflux;

Steps:

5.1.1. 5-Phenyl-1,3,4-thiadiazol-2-amino (2a)

General procedure: A stirring mixture of benzoic acid (6.10 g, 50 mmol), N-aminothiourea (4.55 g, 50 mmol) and POCl3 (13 ml) was heated at 75 °C for 0.5 h. After cooling down to room temperature, water (55 ml) was added. The reaction mixture was refluxed for 4 h. After cooling, the mixture was basified to pH 8 by the dropwise addition of 50% NaOH solution under stirring. The precipitate was filtered and recrystallized from ethanol to yield 8.14 g of the target compound 2a as a colorless crystal. Yield: 92%
