6-Bromoimidazo[1,2-a]pyridine is an intermediate in organic synthesis and pharmaceutical research and development, which can be used in the process of laboratory organic synthesis and chemical and pharmaceutical synthesis. 2-amino-6-bromopyridine , bromoacetaldehyde diethyl acetal and 48 % Hydrobromic acid aqueous solution undergo ring-closure reaction to synthesis 6-bromoimidazo[1,2-a]pyridine.

Reaction Conditions:

with hydrogen bromide in ethanol;water at 100; for 8 h;

Steps:

1 Intermediate Preparation Example 1 6-Bromo-imidazo[1,2-a]pyridine
Dissolve 2-amino-6-bromopyridine (3g, 17.34mmol, 1.0equiv) in a mixed solution of 30mL ethanol and 18mL water, add bromoacetaldehyde diethyl acetal (5.13g, 26.01mmol, 1.5equiv) and 48 % Hydrobromic acid aqueous solution (3mL), after the feeding, the temperature was raised to 100°C for 8h. TLC plate monitors that the reaction is no longer proceeding. Cool the reaction solution to room temperature, concentrate under reduced pressure, add 100mL water, extract with ethyl acetate (100mL×3), combine the organic phases and wash with saturated brine (50mL×3). Drying with sodium sulfate, filtration and concentration under reduced pressure, the residue is separated by column chromatography to obtain 2.98 g of white solid powder, which is the intermediate 6-bromoimidazo[1,2-a]pyridine, the yield is 87%.
