47.1 Step 1: 6-cyano-1H-indole-2-carboxylic acid

To a suspension of methyl 6-cyano-1H-indole-2-carboxylate (1.00 g, 5.00 mmol) in MeOH (15 mL) was added 1 M sodium hydroxide (7.5 mL, 7.49 mmol) at ambient temperature. The mixture was stirred at 40°C for 2 h. After cooling to room temperature, the reaction was acidified with 2M HCl (5 mL) and water (20 mL) was added. The resulting precipitate was collected by vacuum filtration, washed with water (50 mL) and dried in the vacuum oven at 45°C overnight to afford the title compound (926 mg, 95 %) as pale yellow solid. Method A: LC-MS (electrospray): m/z = 185.2 (M-H)-, RT = 0.98 min
