Reaction Conditions:

Stage #1:ethyl thieno<2,3-b>pyrrole-5-carboxylate with sodium hydroxide;ethanol;water for 1 h;Heating / reflux;
Stage #2: with hydrogenchloride;water in ethanol at 20; for 0.5 h;

To a solution (10 ml) of 16 (226 mg, 1.16 mmol, 1 eq.) in ethanol, an aqueous solution (5 ml) of sodium hydroxide (740 mg, 18.5 mmol, 15.9 eq.) was added, and the mixture was stirred for 1 hour under reflux. After allowing the mixture to cool, 6N aqueous hydrochloric acid solution was added. Ethyl acetate was added to the reaction system and the resulting mixture was washed twice with water and once with saturated brine. After drying the resulting mixture over anhydrous sodium sulfate, the mixture was concentrated under reduced pressure to obtain gray solids 17 (190 mg, 97.7%). TLC (silica): Rf = 0.4 (hexane :ethyl acetate = 1 : 1)
