2 Preparation of 3-((dimethylamino)methylene)piperidine-2,4-done (C1)
To a solution of piperidine-2,4-dione (2.00 g, 17.7 mmol) in dimethylformamide (70.7 mL) was added 1,1-dimethoxy-N,N-dimethylmethanamine (2.47 mL, 18.6 mmol). The reaction was stirred at 90° C. for 4 hours, then evaporated to dryness under reduced pressure to give Compound C1 as a brown oil (3.17 g, 85%): 1H NMR (300 MHz, DMSO-d6) δ 7.94 (s, 1H), 3.24 (s, 2H), 3.13 (s, 2H), 2.87 (s, 3H), 2.71 (s, 3H).
