A hydroxyl group at the 4 position of commercially available vanillic acid (Sigma-Aldrich) and a hydroxyl group of carboxylic acid were benzyl-protected in a dimethylformamide solvent (DMF, Thermo Fisher Scientific K.K.) using benzyl bromide (Sigma-Aldrich). Then, the unreacted product was removed using an open column (silica gel 60 (spherical); Kanto Chemical Co., Inc.) (hexane : ethyl acetate = 8:1), followed by purification. The yield was found to be 100%: 1H NMR (300 MHz, CDCl3): δ7.65 (1H, dd, H-6), 7.59 (1H, d, H-2), 7.41-7.26 (9H, m, H-benzylic CH2), 6.88 (1H, d, H-5), 5.33 (2H, s, H-benzylic CH2), 5.21 (1H, s, H-benzylic CH2), and 3.92 (3H, s, CH3).
