Example 25 - The Fisher Esterifcation of amino acids and alcohols [0247] The amino acid was suspended in the alcohol, 50 mole equivalents, and cooled to 000 while stirring. Thionyl chloride, 5 mole equivalents, was added dropwise to the cooled solution. The drop rate was monitored to ensure the temperature does not exceed 1000. Upon completion of the thionyl chlorideaddition, the reaction was heated at 95°C for 5 hours. The reaction mixture was cooled to room temperature, then placed in an ice bath and ethyl ether added until the solution turned turbid. This typically required 5x the volume of alcohol used. The mixture was allowed to set at 000 for an hour and filtered. The crude product was recrystallized from ethanol and ethyl ether.
