Reaction Conditions:

with 1-ethyl-(3-(3-dimethylamino)propyl)-carbodiimide hydrochloride in N,N-dimethyl-formamide at 20; for 16 h;

To a solution of Biotin (127 mg, 0.519 mmol) in DMF (2 mL) was added EDC-HCl (99 mg, 0.519 mmol) and amine (7) (100 mg, 0.433 mmol), and the whole was stirred at rt for 16 h. The reaction mixture was co-evaporated with toluene (x3), followed by purification using flash chromatography (CH2Cl2/MeOH = 5:1) gave desired product S8 (179 g, 90%) as colorless oil. 1H NMR (400 MHz, MeOD): δ 4.49 (dd, J = 8.0, J = 5.0 Hz, 1H), 4.31 (dd, J = 7.9, J = 4.5 Hz, 1H), 4.19 (d, J = 2.5 Hz, 2H), 3.69-3.61 (m, 12H), 3.56 (t, J = 5.3 Hz, 2H), 3.36 (m, 2H), 3.20 (m, 1H), 2.93 (dd, J = 12.8, 5.0 Hz, 1H), 2.85 (t, 1H, J = 2.4 Hz), 2.71 (d, J = 12.7 Hz, 1H), 2.23 (t, J = 7.3 Hz, 2H), 1.77-1.41 (m, 6H).
