Steps:
1 Preparation of 5,5'-dithiobis(succinimidyl-2-nitrobenzoate) (DSNB) Compound
Step (1): 1.3 mmoles of 5,5'-dithiobis(2-nitrobenzoic acid) (DNBA), 2.5 mmole of dicyclohexylcarbodiimide (DCCD), 2.6 mmoles of N-hydroxysuccinimide Amine (NHS) and 20 mL of tetrahydrofuran (THF) to give a mixture.Step (2): The mixture was stirred at 25 to 28 ° C for 2 hours to allow the mixture to react to obtain a reaction solution.Step (3): The reaction solution was filtered to remove the residual solid to give a filtered solution.Step (4): The filtered solution was added to 3 g (24.91 mmole) of magnesium sulfate desiccant, and the filtered solution was dehydrated and dried to obtain a dehydrated solution.Step (5): THF in the dewatered solution was removed by means of reduced pressure concentrating to obtain a crude product as a solid.Step (6): The DSNB compound (white solid, yield 97%) was prepared by dissolving the crude solid with 10 mL of ethyl acetate and then adding 400 mL of n-hexane dropwise under supersonic shaking.
