Preparation by nitration of 2-hydroxy-4-methoxyaceto-phenone (paeonol) with nitric acid (d = 1.42) in acetic acid (47%).

Steps:

PREPARATION OF 1-(2-hydroxy-4-methoxy-5-nitrophenyl)ethanone.

To 25 mL of HN03 (69%) at 0°C, Intermediate 16 (2g, 12mmol) was added and the mixture was kept under constant stirring at the same temperature for 4h. Then the solution was poured onto iced water (200 g) and allowed to precipitate overnight at 4°C. The brown solid formed was collected by filtration to afford 2.2 g (86%); Anal. Calc. for CgHgNOs: C, 51 .19; H, 4.30; N, 6.63. Found: C, 51 .19; H, 4.30; N, 6.63. 1H NMR (300MHz, DMSO-of6) δ 2.62 (s, 3H), 3.96 (s, 3H), 6.78 (s, 1 H), 8.48 (s, 1 H), 12.76 (s, 1 H). 13C NMR (75 MHz, DMSO-ofe) δ 27.13, 57.30,101.61 , 1 13.36, 130.15, 131.56, 158.43, 165.98, 201 .76.
