Synthesis of ethyl 3-amino-4-(methylamino)benzoate (1e)
To a solution of ethyl 4-(methylamino)-3-nitrobenzoate (13.0 g, 58 mmol) in methanol (180 mL) was added slurry of 10% Pd/C (1.3 g in 10 mL of ethanol) under nitrogen atmosphere.
The flask was kept in a Parr shaker at RT under hydrogen pressure (60 psi) for 5 h.
The reaction mixture was filtered through celite bed and concentrated under vacuum to afford the title compound (10.0 g, 88%); 1H NMR (400 MHz, DMSO-d6): δ 7.23 (dd, J=1.6 Hz, J=8.4 Hz 1H), 7.16 (d, J=1.6 Hz, 1H), 6.39 (d, J=8.4 Hz, 1H), 5.38-5.37 (m, 1H), 4.66 (s, 2H), 4.18 (q, J=7.2 Hz, 2H), 2.77 (d, J=5.2 Hz, 3H), 1.26 (t, J=7.2 Hz, 3H); LC-MS: m/z 195.1 (M-1)+.
