Step 1 Synthesis of 6-Pyrrolidin-1-yl-nicotinic acid ethyl ester Potassium carbonate (3.89 g, 28 mmol) was added to a stirred solution of pyrrolidine (1.0 g, 14 mmol) and 6-chloronicotinic acid ethyl ester (2.87 g, 15.5 mmol) in DMF (10 mL) and the resulting mixture was stirred with heating at 60° C. overnight. Cold water was then added to and the resulting precipitate was dried to afford 498 mg (80%) of 6-pyrrolidin-1-yl-nicotinic acid ethyl ester. LCMS-purity: 97%.
