Ethyl 2-diethoxyphosphoryl)acetate (25.0 g, 111.6 mmol, I equiv) was dissolved in deuterium oxide (125 mL, Cambridge isotopes, 99 atom%D) and potassium carbonate was added (150 mg, 1.1 mmol, 0.01 equiv) The reaction mixture was stirred at room temperature for 3 days. The mixture was extracted with ethyl acetate (3 x 150 rnL). The combined organic layers were washed with saturated sodium chloride in deuterium oxide (1 x 100 rnL), dried over sodium sulfate, filtered, and concentrated under reduced pressure to yield ethyl 2- (diethoxyphosphoryl)acetate-d2 (25.0 g, >99% yield) as a colorless oil.
